RESUMO

The growth and development of phytoplankton are influenced by physico-chemical parameters, which can also affect the spatial distribution of phytoplankton community structure. However, it is unclear whether environmental heterogeneity caused by multiple physico-chemical factors can affect the spatial distribution of phytoplankton and its functional groups. In this study, we investigated the seasonal variation and spatial distribution of phytoplankton community structure and its relationships with environmental factors in Lake Chaohu from August 2020 to July 2021. We recorded a total of 190 species from 8 phyla, which were divided into 30 functional groups, including 13 dominating functional groups. The average annual phytoplankton density and biomass were (5.46 ± 7.17) × 107 cells/L and 4.80 ± 4.61 mg/L, respectively. The density and biomass of phytoplankton were higher in summer ((14.64 ± 20.34) × 107 cells/L, 10.61 ± 13.16 mg/L) and autumn ((6.79 ± 3.97) × 107 cells/L, 5.57 ± 2.40 mg/L), with the M and H2 of dominant functional groups. The dominant functional groups were N, C, D, J, MP, H2, and M in spring, whereas functional groups C, N, T, and Y dominated in winter. The distribution of phytoplankton community structure and dominant functional groups exhibited significant spatial heterogeneity in the lake, which was consistent with the environmental heterogeneity of the lake and could be classified into four locations. Location I had higher phytoplankton density and biomass than the other three locations. Additionally, dominant functional groups M, C, and H2 were present throughout the lake, and all 13 dominant functional groups were observed in Location II. Our findings suggest that environmental heterogeneity is a key factor influencing the spatial distribution of phytoplankton functional groups in Lake Chaohu.

BACKGROUND: Molting is an important physiological process in the growth and development of arthropoda, which is mainly regulated by juvenile hormone and ecdysone. CYP302A1 is a key enzyme which plays a critical role in the synthesis of ecdysone in insects, but it has not been identified in cladocera. RESULTS: The CYP302Al gene of Daphnia sinensis was cloned and its function was analyzed in this paper. The CYP302Al gene of D. sinensis was 5926 bp in full-length, with an open reading frame (ORF) of 1596 bp that encoded 531 amino acids (aa), a molecular weight of 60.82 kDa and an isoelectric point of 9.29. The amino acid sequence analysis revealed that there were five characteristic conserved regions of cytochrome P450 family (namely helix-C, helix-K, helix-I, PERF and heme-binding). In dsRNA mediated experiment, the expression level of CYP302A1 gene decreased significantly (knock-down of 56.22%) in the 5% Escherichia coli concentration treatment. In addition, the expression levels of EcR and USP and HR3 genes in the downstream were also significantly decreased, whereas that of FTZ-f1 gene increased significantly. In the 5% E. coli treatment, the molting time at maturity of D. sinensis prolonged, and the development of embryos in the incubation capsule appeared abnormal or disintegrated. The whole-mount in situ hybridization showed that the CYP302A1 gene of D. sinensis had six expression sites before RNA interference (RNAi), which located in the first antennal ganglion, ovary, cecae, olfactory hair, thoracic limb and tail spine. However, the expression signal of the CYP302A1 gene of D. sinensis disappeared in the first antennal ganglion and obviously attenuated in the ovary after RNAi. CONCLUSION: The CYP302A1 gene played an important role in the ecdysone synthesis pathway of D. sinensis, and the knock-down of the gene affected the molting and reproduction of D. sinensis.

BACKGROUND: In the life history of Daphnia, the reproductive mode of parthenogenesis and sexual reproduction alternate in aquatic ecosystem, which are often affected by environmental and genetic factors. Recently, the sex-biased genes are of great significance for clarifying the origin and evolution of reproductive transformation and the molecular regulation mechanism of sex determination in Daphnia. Although some genes on reproductive transition of Daphnia had been researched, molecular mechanism on the maintenance of sexually dimorphic phenotypes of Daphnia are still not well known, including differentially expressed genes in different life-history stages. RESULTS: In this study, four life-history stages of Daphnia sinensis, juvenile female (JF), parthenogenetic female (PF), sexual female (SF) and male (M), were performed for transcriptome, and male-biased genes were screened. A total of 110437 transcripts were obtained and assembled into 22996 unigenes. In the four life-history stages (JF, PF, SF and M), the number of unique unigenes is respectively 2863, 445, 437 and 586, and the number of common unigenes is 9708. The differentially expressed genes (DEGs) between male and other three female stages (M vs JF, M vs PF and M vs SF) were 4570, 4358 and 2855, respectively. GO gene enrichment analysis showed that the up-regulated genes in male were mainly enriched in hydrolase activity and peptidase activity. Thirty-six genes in male were significantly higher expression than in the three female stages, including one Doublesex (Dsx) gene, one laminin gene, five trypsin genes and one serine protease genes, and one chitin synthase gene and two chitinase genes. CONCLUSIONS: Our results showed that thirty-six candidate genes may be as the male-biased genes involving in the maintenance of sexually dimorphic phenotypes. This work will provide a reference for further exploring the functional genes related to sex differentiation in Daphnia species. Moreover, according to previous investigations, we thought that the expression level of functional genes may be related to the life-history stages of organisms, and may be also affected by different Daphnia species.

Lake eutrophication and cyanobacterial blooms have become worldwide environmental issues. Under cyanobacterial blooms (especially Microcystis), Daphnia spp. can transfer beneficial information to their offspring in order to improve adaptability. Hox genes are important regulatory factors of transcription in metazoans, and are involved in the growth and development of organisms. However, the mechanisms of Microcystis on the expression of Hox genes in Daphnia are unclear. In this study, the effects of Microcystis aeruginosa on Hox gene expression in the mothers and offspring (F1) of two Daphnia similoides sinensis clones were investigated using a mixed diet of M. aeruginosa and Scenedesmus obliquus. Compared with the 100%S food treatment, the survival rates at the end of the experiment of clone 1-F1 in the food treatments containing M. aeruginosa were significantly lower, but it was significantly higher for clone 2-F1 in the 20%M + 80%S food treatment. Moreover, the survival rates at the end of the experiment of clone 1-F1 in the food treatments containing M. aeruginosa were significantly higher than those of their mother. Based on previous transcriptome data, 14 Hox genes of D. similoides sinensis were identified, including Abd-B, CDX-1, Dll, HOX-1, HOX-2, HOXA1, HOXA2, HOXB3, HOXB3-2, HOXB7, HOXC4, HOXC7, HOXC8, and HOXD10. The expressions of Abd-B, HOX-2, HOXA1, HOXC7, and HOXD10 of clone 2-mothers in the 40%M + 60%S food treatment were 2.9-22.5 times as high as in the 100%S food treatment, whereas the expressions of CDX-1, HOX-1, HOXB3, and HOXD10 of clone 1-mothers were 4.8-13.1 times at same food level. The expression of HOXA2, HOXC7, HOXC8, and HOXD10 of clone 1-F1 in the 40%M + 60%S food treatment was 8.2-21.1 times as high as in the 100%S food treatment. However, compared with the 100%S food treatment, the expressions of CDX-1 in the mothers and F1 of clone 2 and HOXB7 in the mothers of clone 1 in the food treatments containing M. aeruginosa were significantly lower (p < .05). Our results suggest that the offspring (F1) produced by D. similoides sinensis mother pre-exposed to toxic M. aeruginosa had stronger adaptability to M. aeruginosa than their mothers. Moreover, Hox gene expressions of D. similoides sinensis had obvious differences between clones under stress of toxic M. aeruginosa.

Cladoceran remains (e.g., Daphnia ephippia) in sediments are usually able to accurately reflect the historical succession of cladoceran in lakes. However, mechanisms describing the interaction between density changes of empty ephippia and ephippia containing resting eggs in lake sediments and environmental factors remain unclear. The diversity and vertical changes of Daphnia ephippia in the 30-cm sedimentary layer (equivalent to about 90 years) were investigated in a subtropical Chinese lake, Lake Chaohu. The ephippia of D. similoides sinensis, D. pulex, and D. galeata were identified in the lake sediments. The densities and accumulation rates of both empty ephippia and ephippia containing resting eggs of three Daphnia species showed similar patterns. The values in the surface sedimentary layers (after the 1970s) were notably higher than in the bottom sedimentary layers (before the 1970s). The densities and accumulation rates of both empty ephippia and ephippia containing resting eggs of D. similoides sinensis were higher than those of D. pulex and D. galeata. Highly significant relationships (P < 0.001) between the TN contents and the ephippial densities and accumulation rates of D. similoides sinensis and D. galeata. TP contents had also significant correlations with the ephippial densities and accumulation rates of D. similoides sinensis and D. galeata, whereas it was not significant correlations with those of D. pulex (P > 0.05). Our results suggested that long-term eutrophication and cyanobacterial blooms might significantly affect the production of Daphnia ephippia and their vertical distribution in the sediments in eutrophic lakes.

Mitochondrial 16S rDNA and CO I gene were used as molecular markers for the analysis of the genetic diversity and differentiation of Daphnia galeata populations in nine water bodies in the middle and lower reaches of the Yangtze River. In the combined 16S rDNA and CO I gene sequences, 54 variable sites and 44 haplotypes were observed among 219 individuals belonging to nine D. galeata populations. Average haplotype diversity and nucleotide diversity were, respectively, 0.72% and 0.56%. The F-statistics (F ST) value of the D. galeata populations was 0.149. According to the results of the neutral test, D. galeata in the middle and lower reaches of the Yangtze River had experienced a bottleneck effect in the history. Molecular variance analysis indicated that the genetic differentiation of the D. galeata populations mainly occurred within populations (85.09%). Greater genetic differentiations of D. galeata among individuals within populations appeared in the populations from the Huaihe River basin, whereas smaller genetic differentiations occurred in the populations from the middle reaches of the Yangtze River. Strong gene flows were all observed between Group I (four populations from the middle reaches of the Yangtze River) and Group ΙΙ (three populations from the middle and lower reaches of the Yangtze River), and Group ΙΙΙ (two populations from the Huaihe River basin). The effective migration rates (M) were 851.49 from Group I to Group ΙΙ and 685.96 from Group I to Group ΙΙΙ, respectively. However, no significant relationship was observed between the genetic differentiation and geographical distance of the nine populations (r = .137, p > .05). Results suggested that the genetic differentiation of D. galeata in the water bodies in the middle and lower reaches of the Yangtze River resulted mainly from geographical isolation.

Geographical patterns, climate, and environmental change have important influences on the distribution and spread of aquatic organisms. However, the relationships between the geographical pattern and phylogenetics of Daphnia as well as environmental change are not well known. The genetic diversity and phylogeography of seven D. similoides sinensis populations located in the middle and lower reaches of the Yangtze River were investigated based on the combination of mitochondrial (COI gene) and nuclear (14 microsatellite primers) markers. Based on the mitochondrial gene markers, D. similoides sinensis from the middle and lower reaches of the Yangtze River had one ancestral haplotype and two evolutionary clades. In addition, D. similoides sinensis population deviated from neutral evolution, showing signs of a bottleneck effect followed by population expansion. Based on the microsatellite markers, the seven D. similoides sinensis populations formed three main groups. The dendrogram (NJ/ME) showed that D. similoides sinensis based on the mitochondrial genes marker were obviously clustered two main clades, whereas there were three clades based on the microsatellite markers. Our results suggested that the habitat fragmentation due to the barrier of the dams and sluices promoted the genetic differentiation and phylogeography of D. similoides sinensis populations in the middle and lower reaches of the Yangtze River.

With water eutrophication and global warming, cyanobacteria blooms have occurred frequently, and the interaction between M. aeruginosa and Daphnia has been widely paid attention by researchers. However, the effects of toxic M. aeruginosa on the SOD activity of Daphnia are poorly known. Six D. similoides sinensis clones collected from Lake Junshan and the offspring of two clones were employed. The effects of toxic M. aeruginosa on the life history traits and SOD activities of D. similoides sinensis in the mother and their offspring were studied. Toxic M. aeruginosa could significantly inhibit the life history traits (e.g., body lengths, offspring numbers at first reproduction, cumulative offspring numbers, and the intrinsic rate of population) and induce higher SOD activities of D. similoides sinensis. Compared with the mother, the effects of toxic M. aeruginosa on the life history traits and SOD activities of D. similoides sinensis in the offspring showed obvious differences. Moreover, the adaptability of the offspring to M. aeruginosa indicated also the differences between two clones. Our results suggested that the mother exposed to toxic M. aeruginosa could enhance the fitness of their offspring to Microcystis by maternal effect and was also affected by the D. similoides sinensis genotypes.

Nuclear receptor (NR) genes form a conserved superfamily, which is involved in organism metabolism, reproduction, development, homeostasis, and resource allocation. Microcystis aeruginosa can inhibit the growth and reproduction of Daphnia. However, whether M. aeruginosa can affect the expression of Daphnia NR genes is unknown. In total, 18 NRs were identified in this study based on previous Daphnia similoides sinensis transcriptome data. In treatments containing M. aeruginosa, the gene expression of the NR1 subfamily (E75a, E75b, HR3, HR96, NHR-1, HR97a, HR97g, and NHR97) and the NR2 subfamily (RXR, TLL, PNR, and SVP) were down-regulated 59% and 79%, respectively. In treatments containing M. aeruginosa, although the expression of 78% of the genes showed a similar trend in clones 1 and 2, the expression of 42% of the genes in clone 3 showed the opposite trend compared to clones 1 and 2, suggesting that the adaptability and molecular mechanism differ in individuals with different Microcystis tolerance genotypes.

Daphnia pulex has played an important role in fresh-water ecosystems. In this study, the complete mitochondrial genome of Daphnia pulex from Chaohu, China was sequenced for the first time. It was accomplished using long-PCR methods and a primer-walking sequencing strategy with genus-specific primers. The mitogenome was found to be 15,306 bp in length. It contained 13 protein-coding genes, two rRNA genes, 22 tRNA genes and a typical control region. This research revealed an overall A+T content of 64.50%. All of the 22 typical animal tRNA genes had a classical clover-leaf structure except for trnS1, in which its DHU arm simply formed a loop. The lengths of small and large rRNA were 744 bp and 1,313 bp, respectively. The A+T-rich region was 723 bp in length, which is longer than that from the North American species (689 bp). In terms of structure and composition, many similarities were found between the Chinese and North American Daphnia pulex.

The water flea Daphnia are planktonic crustaceans commonly found in freshwater environment that can switch their reproduction mode from parthenogenesis to sexual reproduction to adapt to the external environment. As such, Daphnia are great model organisms to study the mechanism of reproductive switching, the underlying mechanism of reproduction and development in cladocerans and other animals. However, little is known about the Daphnia's reproductive behaviour at a molecular level. We constructed a genetic database of the genes expressed in a sexual female (SF) and a parthenogenetic female (PF) of D. similoides using Illumina HiSeq 2500. A total of 1,763 differentially expressed genes (865 up- and 898 down-regulated) were detected in SF. Of the top 30 up-regulated SF unigenes, the top 4 unigenes belonged to the Chitin_bind_4 family. In contrast, of the top down-regulated SF unigenes, the top 3 unigenes belonged to the Vitellogenin_N family. This is the first study to indicate genes that may have a crucial role in reproductive switching of D. similoides, which could be used as candidate genes for further functional studies. Thus, this study provides a rich resource for investigation and elucidation of reproductive switching in D. similoides.

Chemosensory receptors play key roles in insect behavior. Thus, genes encoding these receptors have great potential for use in integrated pest management. The hover fly Scaeva pyrastri (L.) is an important pollinating insect and a natural enemy of aphids, mainly distributed in the Palearctic and Nearctic regions. However, a systematic identification of their chemosensory receptor genes in the antennae has not been reported. In the present study, we assembled the antennal transcriptome of S. pyrastri by using Illumina sequencing technology. Analysis of the transcriptome data identified 60 candidate chemosensory genes, including 38 for odorant receptors (ORs), 16 for ionotropic receptors (IRs), and 6 for gustatory receptors (GRs). The numbers are similar to those of other Diptera species, suggesting that we were able to successfully identify S. pyrastri chemosensory genes. We analyzed the expression patterns of all genes by using reverse transcriptase PCR (RT-PCR), and found that some genes exhibited sex-biased or sex-specific expression. These candidate chemosensory genes and their tissue expression profiles provide information for further studies aimed at fully understanding the molecular basis behind chemoreception-related behaviors in S. pyrastri.

Increased human activities and environmental changes may lead to genetic diversity variations of Cladocerans in water. Daphnia pulex are distributed throughout the world and often regarded as a model organism. The 16S rDNA, cytochrome c oxidase subunit I (COI), and 18S genes were used as molecular marks. The genetic diversity and phylogeny of D. pulex obtained from 10 water bodies in the middle and lower reaches of the Yangtze River were studied. For 16S rDNA, COI gene, and 18S gene, the A+T content (65.4%, 58.4%, and 54.6%) was significantly higher than the G+C content (34.6%, 41.6% and 45.4%). This result was consistent with higher A and T contents among invertebrates. Based on the genetic distances of 16S rDNA and COI genes, the genetic differences of D. pulex from 10 water bodies located in the middle and lower reaches of the Yangtze River in China was minimal (0%-0.8% for 16S rDNA and 0%-1.5% for COI gene). However, D. pulex evolved into two branches in the phylogenetic trees, which coincided with its geographical distribution. Compared with D. pulex from other countries, the average genetic distance of D. pulex obtained from 10 water bodies in the middle and lower reaches of the Yangtze River reached 9.1%-10.5%, thereby indicating that D. pulex may have evolved into different subspecies.

Shengjin Lake is an important wintering and stopover habitat for migratory birds en route from East Asia to Australia. The authors investigated the rotifer community structure and environmental factors in upper Shengjin Lake from February to July of 2014. A total of 17 genera and 31 species of rotifers were identified, the dominant species being Brachionus angularis, Brachionus calyciflorus, Asplanchna priodonta, and Schizocerca diversicornis. Rotifer density and the nutrient salt content were higher in the areas inhabited by migratory birds. The highest density (176.0 ind./L) was recorded in June, the lowest (14.0 ind./L) in April. Canonical correspondence analysis (CCA) and correlation analysis showed that some environmental factors, such as temperature, pH, water depth, Secchi depth (SD), and chlorophyll a concentration, had a considerable effect on the lake's rotifer community structure.

In this paper, we determined the complete mitochondrial genome of Chinese Daphnia carinata for the first time by the long and accurate polymerase chain reaction and primer-walking methods. It was 15,245 bp in length, with an A + T content of 70.35%, containing 37 typical animal mitochondrial genes and an A + T-rich region. The COI gene started with ACTA. All the 22 typical tRNA genes had a classical cloverleaf structure except for trnS1, in which the D-stem pairings in the DHU arm were absent.

BACKGROUND: Since chemosensory genes play key roles in insect behaviour, they can potentially be used as new targets for pest control. The cabbage beetle, Colaphellus bowringi, is a serious insect pest of cruciferous vegetables in China and other Asian countries. However, a systematic identification of the chemosensory genes expressed in the antennae has not been reported. RESULTS: We assembled the antennal transcriptome of C. bowringi by using Illumina sequencing technology and identified 104 candidate chemosensory genes by analyzing transcriptomic data, which included transcripts encoding 26 odorant-binding proteins (OBPs), 12 chemosensory proteins (CSPs), four sensory neuron membrane proteins (SNMPs), 43 odorant receptors (ORs), nine ionotropic receptors (IRs), and ten gustatory receptors (GRs). The data obtained are similar to those found in other coleopteran species, suggesting that our approach successfully identified the chemosensory genes of C. bowringi. The expression patterns of 43 OR genes, some of which were predominately found in the antenna or associated with sex-biased expression, were analyzed using quantitative real time RT-PCR (qPCR). CONCLUSIONS: Our study revealed that a large number of chemosensory genes are expressed in C. bowringi. These candidate chemosensory genes and their expression profiles in various tissues provide further information on understanding their function in C. bowringi as well as other insects, and identifying potential targets to disrupt the odorant system in C. bowringi so that new methods for pest management can be developed.

Although the general pathway of sex pheromone synthesis in moth species has been established, the molecular mechanisms remain poorly understood. The common cutworm Spodoptera litura is an important agricultural pest worldwide and causes huge economic losses annually. The female sex pheromone of S. litura comprises Z9,E11-14:OAc, Z9,E12-14:OAc, Z9-14:OAc, and E11-14:OAc. By sequencing and analyzing the transcriptomic data of the sex pheromone glands, we identified 94 candidate genes related to pheromone biosynthesis (55 genes) or chemoreception (39 genes). Gene expression patterns and phylogenetic analysis revealed that two desaturase genes (SlitDes5 and SlitDes11) and one fatty acyl reductase gene (SlitFAR3) showed pheromone gland (PG) biased or specific expression, and clustered with genes known to be involved in pheromone synthesis in other moth species. Furthermore, 4 chemoreception related genes (SlitOBP6, SlitOBP11, SlitCSP3, and SlitCSP14) also showed higher expression in the PG, and could be additional candidate genes involved in sex pheromone transport. This study provides the first solid background information that should facilitate further elucidation of sex pheromone biosynthesis and transport, and indicates potential targets to disrupt sexual communication in S. litura for a novel pest management strategy.

Microcystis colonial size and concentration have detrimental effects on life history traits of Daphnia, but their detailed interactions have remained unclear so far. Our experiments show that the interaction between Microcystis colonial size and concentration on maturation time, life expectancy, net reproductive rate and innate capacity of increase in Daphnia similoides was significant. In all groups, the survival rate of D. similoides was 100% within 8 days. This value then declined quickly in the large-colony groups and in the SH group of Microcystis. Colonial M. aeruginosa significantly reduced the maturation time and body length at maturity of D. similoides. The number of offspring at first reproduction per female in the SH group of Microcystis was significantly higher than those in other groups. Net reproductive rate of D. similoides in the SL group of Microcystis was significantly higher than those in other groups of Microcystis. The innate capacity of increase of D. similoides in small-colony Microcystis groups was significantly higher than that in the large-colony groups. The results suggested that the effect of smallcolony Microcystis on the reproduction of Daphnia was positive under lower concentration, while their toxicity was intensitied under higher concentration when small-colony Microcystis were by Daphnia as food.

Temporal and spatial variations in the phytoplankton community and environmental variables were investigated from February to July 2014, in the upper lake of Shengjin Lake, China. We identified 192 species of phytoplankton belonging to 8 phyla and 84 genera, of which 46.4% of Chlorophyta, 29.2% of Bacillariophyta, and 12.5% of Cyanophyta. There were 14 predominant species. Marked temporal and spatial variations were observed in the phytoplankton community. The total abundance of phytoplankton ranged from 3.66 × 10(5) to 867.93 × 10(5) cells/L and total biomass ranging from 0.40 to 20.89 mg/L. The Shannon-Wiener diversity index varied from 3.50 to 8.35 with an average of 5.58, revealing high biodiversity in the phytoplankton community. There were substantial temporal changes in the dominant species, from Bacillariophyta and Cryptophyta to Cyanophyta and Chlorophyta. Phytoplankton biomass and abundance showed a similar increasing trend from February to July. Pearson correlations and Redundancy analysis revealed that the most significant environmental factors influencing phytoplankton community were water temperature (T), transparency (SD), and nutrient concentration. The positive correlation between the key water bird areas and phytoplankton biomass indicated that the droppings of wintering water birds had an important influence on the phytoplankton community in the upper lake of Shengjin Lake.

A laboratory experiment was conducted to study the effects of different population density (D1 : 100 ind x L(-1), D2 : 150 ind x L(-1), D3 : 300 ind x L(-1)) and culture volume (V1: 50 mL, V2 : 100 mL, V3 : 400 mL) on the growth and reproduction of Moina irrasa at 25 degrees C. At the same culture density, the body length of the M. irrasa females at their first pregnancy, the first brood, and the total offsprings per female decreased with the increase of culture volumes, while the sex ratio (male/female) of the offsprings was in adverse. At the same culture volumes, the total offsprings per female decreased with the increase of culture density. At D1 V1, the body length of the females at their first pregnancy (0.95 +/- 0.10 mm) and the total offsprings (171.3 +/- 19.8 ind) per female were the maximum. At D3V2, the sex ratio was the maximum (0.54 +/- 0.05). Culture density, culture volume, and their interactions significantly affected the total offsprings per female and the sex ratio (P < 0.001).
